The connection from Mara to Serengeti (and v.v.), is offered in conjunction with Coastal Aviation and is accessible via Migori (on the Kenyan side) and Tarime (on the Tanzanian side) near the border point of Isabania. The connection involves a short scenic road transfer between the two strips of Migori and Tarime, it is a convenient and time saving option for clients wanting to experience both the Serengeti and Masai Mara.
The service commenced in January 2012 and operates during the high seasons only from Mid December to end March and beginning of July to end of October each year. During these periods, the service operates daily subject to minimum 2 pax per strip, starting from the Tanzanian side with Coastal Aviation picking clients from the various strips in Serengeti (according to number of pax booked) from approximately 0820 and arriving at Tarime by 1025. There is a road transfer via Isabania border point for immigration clearance, before proceeding to Migori on the Kenyan side, where clients will connect onto the Safarilink flight to Masai Mara departing at 1215.
The reverse of this schedule from Masai Mara to Serengeti departs the Mara at 1130 to arrive at Migori at 1200 where clients are met for the road transfer to Tarime via Isabania. Arrival at Tarime is approximately 1330 to meet the Coastal aircraft that departs at 1400 for the Serengeti destinations.

Además de carretera (en este caso camino) necesitarías que hubiese puesto fronterizo donde hacer los trámites de entrada o salida de ambos países.
Si no me equivoco el paso más cercano a Masai Mara-Serengeti es Isibania-Sirari, cerca del lago Victoria.

Puedo contestar bien esta pregunta porque el año pasado yo fui de Masai Mara al Seregueti. Fui por libre, con un 4x4 alquilado y 4 personas más. Y la ruta fue de lo más improvisada. Especialmente la decisión a última hora de hacer esto que te explico a continuación:
Como ya sabreís no existe forma legal de cruzar entre un parque i otro por la zona que sería más normal, dado que están separados por la frontera keniata/tanzana. Hay que dar una vuelta. No hubo forma de saltarse esta norma, salvo que cruzaramos ilegalmente lo cual comportaría un grave problema de papeles en Tanzania, donde a menudo te para la poli. Por controles. Nosotros salimos por una puerta norte del parque de Masai mara (creo que musiara) y fuimos subiendo por los montes de la "escarpatura del Olooloro". Cuando lleguéis a la cima (que tampoco es muy alto) tendréis una esplendida vista sobre todo Masai mara. Nostros hizimos una buena parada en lo alto de estos montes.
De ahí seguimos por una "carretera" sin asfaltar que según el tramo era mejor o peor, pero con muchos baches. Tuvimos que parar a dormir en una "ciudad" pequeña, Kihancha, en un hotel "de mala muerte" sin agua corriente, (había un pozo de donde sacaron unos cubos de agua) pero fue posible. Al día siguiente, tras más baches de carretera sin asfaltar, llegamos finalmente a la carretera asfaltada A1 . Y de allí rapidamente llegamos al puesto fronterizo de Isebania/Sirari. Una vez hechos los papeles para cruzar, (1/2h como mínimo) seguimos con carretera asfaltada por Tanzania hasta Bunda (muy cerca del lago victoria), y tras haber cruzado el puente que cruza el rio Mara cuando ya está a punto de desembocar en el lago, y es muy ancho, paramos en Bunda, que es una ciudad curiosa. Y poco después encontrámos una entrada al P.N. Serengueti. Y ese mismo día, ya de noche llegamos al campo base de seronera. Pero supuso todo un día casi sin parar de trayecto en 4x4. Claro que cruzando la zona oeste del serengueti (grumeti) ya vimos algunos animales .
Seronera es un campo base que en agosto vimos que tenía como unos 500 campistas de todo el mundo en unas 200 tiendas todas particulares. Había multitud de 4x4. Luego durante el día se veía muy poca presencia humana, pues los 4x4 se dispersan.
Al día siguiente continuámos dentro del parque del serengueti en dirección este. Cruzamos a la area de conservación del ngorongoro...
